I have created a new Studio TM for my project and have DOCX files with source and target in two columns that I want to create a Studio TM of. How can I do that?
You can see how the file layout is here (two columns but also a lot of tags and a heading etc.):

Probably there are dozens of different/better methods, but I would:
1) Copy the 2 columns in Excel; 2) Save the file as tab delimited CSV; 3) Import the CSV in Xbench; 4) Export as TMX; 5) Import the TMX in the Studio TM you created.

that's nice and a kind of standard procedure.
though
If I knew the name of source and target languages, I could reduce the above steps in 2.
1) select an Excel file using converter (then exported as an TMX file) 2) import into Trados
for instance.. "en-US" "ZH-CN" "ko-KR" "JA"...
