A powerful telomerase activator capable of 300% increases in telomerase expression. TA-65 has been shown in mice and human studies to lengthen short telomeres and reverse key biomarkers of aging.
Overview
Telomeres are non-encoding DNA repeats at the tips of the chromosome. In the picture below they are the little yellow dots:
As our cells divide these tips shorten, typically from 12-15 'kilobase pairs' (kb) to 4kb over a lifetime1, the cells which they inhabit become older and more dysfunctional and reproduce less often2. Finally they hit the ‘Hayflick Limit’ at which point they become ‘senescent' and are unable to divide.
Over a decade ago it was discovered that an enzyme called telomerase can lengthen shortening telomeres. It exists in the body's immortal cells - germ cells and embryonic stem cells - but is not expressed in other cell types in quantities great enough to stop the erosion in them. It was shown that by activating telomerase in these types of cells in vitro they become immortalized, do not age across divisions and divide indefinitely.3 After a long search, a compound was discovered that can activate telomerase, in both mice and humans, both in vitro and in vivo, at substantial levels above 300% of normal4. That compound is TA-65.
A New Era
In our opinion aging is by design and telomere shortening is the mechanism by which that design is carried out. If you agree with us on that, you would probably consider it remarkable that for thousands of years, there has never been a way to lengthen them. As our ancestors labored in the creation of elixirs from alchemy and myth, and as the Egyptian, Roman and countless other empires rose and fell, the telomeres of every living human being on Earth moved inexorably in one direction – down.
Amazingly, those days have ended with this product. For the first time in human history we can - with a simple pill - reverse the process that lies at what we believe to be at the bottom of the cascade of cause and effect that is aging. The Forefoot & Hoof
The forefoot of the horse is a product of evolution and a miracle in bioengineering. It is extremely light in weight, flexible and yet it withstands constant shock and concussion due to the forces of locomotion.
The hoof is a highly specialised horny shell composed of three layers. The hoof covers the sensitive bones, nerves, connective tissue and vascular tissue of the forefoot. The primary function of the hoof is to protect the underlying structures, and to increase the surface area for the dissipation of locomotor forces. Hoof growth originates from the coronary band, whereas tissue loss due to friction and abrasive forces occurs on the ground surface of the hoof. Hence, there is unrelenting wear and tear to horses' hooves. Throughout evolution, the horse has adapted to environmental changes. This has led to slow and subtle changes in the appendicular skeleton of the species.
The maintenance of the hoof is dependent on two factors:
1) Nutrition and 2) Moisture content. The hoof horn is composed of cellular tissue (horn cells) with a non-cellular proteinaceous matrix. Keratin Sulphate forms the structural component in this matrix. Keratin sulphate is a mucopolysaccharide, and as such, has a great affinity for water molecules. The integrity of keratin sulphate is fundamental, because keratins' attraction for water facilitates the high moisture content in the hoof horn. The maintenance of moisture content in the hoof horn is extremely important. Elasticity and the ability to absorb compression, are essential factors in the dissipation of the concussive forces of locomotion. If there is inadequate moisture within the hoof, the hoof horn will break down. As already mentioned above, the hoof horn is primarily made from horn cells and keratin sulphate (a proteinaceous material with a high sulphur content). If the hoof horn is somehow deprived of good quality protein, bioavailable sulphur, calcium, zinc or biotin, irregularities can occur in the hoof wall. This will manifest itself as cracks or brittle crumbling hooves. As a species, the horse has evolved and adapted to change well, that is of course until the introduction of man and equestrian sport. Since their domestication, horses have been harnessed to perform work.
Environmental factors such as synthetic working and racing surfaces, roadways, certain bedding and high-grain rations have all put increased pressure on the self-maintenance of horses hooves. In many instances the diet of the stabled horse is not sufficient to meet the nutritional requirements for keratinisation in the beleaguered hoof horn. BIOTEX-37 provides all of the scientifically proven nutrients, which are required by the horse, to maintain healthy hoof production.
Topographically, the hoof is divided into three regions: the toe, the medial and lateral quarters, and the heels. The heels of the wall are reflected inward and form the bars, which are separated from the frog by the paracuneal grooves. The wall is thickest at the toe and gradually thins towards the heel.
The Hoof Wall Structure
The wall is designed to carry the bulk of the horse's weight as well as protect the underlying structures.
The bars act as a brace to control over expansion and contraction of the hoof.
The sole covers the softer tissues and is somewhat concave to give grip and allow for expansion.
The frog aids in absorbing concussion, circulation and regulating moisture in the hoof.
The wall is divided into three layers. The stratum externum, the stratum medium and the stratum internum. When there is a
crack or malformation in the hoof wall, the weakness can normally be located, either in the layer of the wall (stratum externum) or the middle and inner layers (stratum medium and stratum internum). An accurate diagnosis can only be made by electron microscopy and biopsy. While it is impossible to ascertain by the human eye where the weakness originates from, the correct diagnosis is very important in terms of the nutritional therapy that should be applied, to help resolve the problem.
Hoof Growth & The Seasons
The hoof wall itself is composed mainly of keratinised tissue representing the modified epidermis of the skin. However the innermost layer of horn adjacent to the corium, is a growing layer consisting of horn cells. Cells from this layer are continually dividing to produce horn cells which are added to the bulk of the horn. Growth occurs in a tubular fashion from the surface of papillae projecting from the coronary corium.
The hoof wall is therefore formed of tubules of horn tissue held together by non-tubular horn. The direction of growth of horn is downward from the coronet so that tubular horn is pushed downward to produce the wall and bearing surface: thus the youngest horn is closest to the coronary hand, and the oldest horn is on the bearing surface of the wall. This method of growth of the wall explains the presence of ridges in the horn.
When the rate of growth is irregular, alternating ridges and depressions encircle the wall. The nutritional content of the diet, which is affected by the seasons may be an influential factor in the rate of horn production.1 Hoof growth is faster and more prominent in the Spring when the horse is out at grass, producing raised 'grass rings'. During this time, the nutritional content of pasture is significantly than in the Autumn and Winter months. On the other hand, during Winter, hoof growth tends to be diminished and lessened in extent, thus circular depressions can mark the season. The vast majority of horses in training have limited access to quality fresh forage, therefore nutritional deficiencies are not uncommon.
Keratinisation ( Hoof Growth)
Malnutrition will affect physiological processes in the body. These nutritional deficiencies will manifest themselves in many different ways. Irregularities in the epidermal layers, such as the hair and hooves are indicative of nutritional deficiencies in the diet of the horse.
The hoof wall is made up of microscopically sized tubules. The spaces between these tubules are filled with solidly packed horn cells. In this way the tubules are bound together. Keratin is the protein essential to the bonding of these cells and tubules.
This structure allows the horn to flex and absorb concussion, thus reducing wear and tear on the bones, ligaments and tendons of the forefoot. The bulk of the hoof wall, is produced by the coronary band, due to its high cellular content. The coronary band is a strip of highly vascular tissue, which runs around the top of the wall just below the hairline, feet produces horn in much the same way as skin does hair. Horn like hair is primarily protein. Keratinisation is dependant on the normal activity of the horn cells. As there are a high concentration of horn cells in the coronary band, the health of this vascular tissue is fundamental. Nutritional deficiencies, trauma, abrasion, and infection of the coronary band are the most likely limiting factors in the production of keratin in the hoof horn.
Keratinisation is dependant on the healthy activity of horn cells, which need certain nutrients to produce this hoof horn 'adhesive", BIOTEX-37 has been specially formulated to supply a concentrated and balanced level of these nutrients for the optimal rate of keratinisation. This means that BIOTEX-37 will help to control and correct hoof irregularities in all three layers of the hoof wall.
Hoof Defects:

Stratum Externum Stratum Medium Stratum Internum
These three layers arise from the epithelium covering the coronary dermas. The outer layer is an extremely thin layer of tubular horn covering the entire surface of the wall. The middle layer, or stratum medium consists of a very thick hard layer of tubular and inter-tubular horn. The proximal portion divides to form the coronary groove, in which lies the convex coronary corium. It is pigmented when the hoof is dark. The middle portion actually comprises the bulk of the hoof wall. The spaces between the horn tubules are filled with inter-tubular horns. The internal layer or stratum internum is always non-pigmented and consists of many keratinised primary epidermal lamellae which extend from the coronary groove to the ground surface.
Scientific research has found that hoof cracks originate in one or two of these three layers. Research has also shown that the origin of the hoof defect should ultimately decide the choice of nutritional therapy. Cracks that originate in the stratum externum respond very well to biotin supplementation.2 Cracks that originate in the stratum medium and stratum internum will respond better to the addition of protein, calcium, bio-available sulphur and zinc to the biotin supplement.3

Foot Care
A Three Fold Approach
1. Nutrition: All horses in training should receive a balanced diet, if they are expected to perform at the very highest level.
Nourishment of the hoof and the underlying structures should receive primary attention, as they are the foundation of the horse. Lameness is the biggest reason for under performance in the horse. Interestingly, foot lameness is the most common type of lameness seen in the equine athlete.4 BIOTEX-37 is the ultimate feed supplement for hooves, because it provides essential nutrients to nourish every tissue in the hoof horn.
2. Foot Hygiene: First and foremost, stables should be designed to incorporate a 'run-off for urine and water. Fresh air should he a priority in the stable environment. The choice of bedding is subjective. however it is important that whatever bedding used, it should not be too dry as this will compromise the natural moisture content of the hoof horn. The bedding should be kept as fresh as possible. This will reduce the build up and establishment of endemic bacteria and fungi within the stable environment. Several of these micro-organisms have a natural tropism for hoof tissue e.g. Thrush.
3. Farriery: A qualified farrier should always conduct trimming and shoeing work. The farrier has a huge influence on the balance of the foot. Preparation of the hoof, the type of shoe used and its placement, will all affect the displacement of force on the forefoot and adjoining limb. Many hoof defects are multi-factorial in their origin. Incorrect shoeing and neglect can be one of the pre-disposing factors to disease and the breakdown of hoof horn function, cracks and brittle hooves. Proper hoof care, hygienic housing conditions and the feeding of BIOTEX-37 combined, are the best approach for ensuring hoof health and optimum hoof growth.
How does BIOTEX-37 work?
Biotin
Biotin is a member of the B vitamin family yet historically, biotin was referred to as vitamin H. Biotin is a sulphur containing vitamin. Green pasture is the best source of dietary biotin. Horses' also get a supply of biotin and other B vitamins as a by-product of bacterial activity in the hindgut. However, modern husbandry of the performance horse, necessitates large periods of time with little or no access to good quality pasture.
Horses in training fed a high grain / low forage diet can often suffer a B vitamin deficit due to the fact that rapid starch digestion will reduce the natural pH in the caecum. This net result being the depletion of the B vitamin-producing bacteria in the hindgut. Therefore, all stabled horses in training are potentially at risk of a biotin deficiency.
Horses at risk of a biotin deficiency:
• Lack of biotin in the diet.
• Loss of microbial gut activity, from antibiotic therapy or altered gut motility.
• Pregnant and lactating mares, or aged geriatric horses.
• Inherited diseases of biotin metabolism.
Biotin Supplementation
The recommended daily allowance (RDA) of biotin for a 500kg horse is 2 mg per day.5 However, research has shown that increased supplementation of biotin in the diet of horses with hoof problems resulted in improved hoof growth and overall hoof integrity. Every 20g of BIOTEX-37 contains 37.5mg of biotin. This is a concentrated level which maximises biotin absorption in the small intestine. Biotin supplementation will enhance the repair of hoof defects and. will help prevent their recurrence. There is good scientific evidence that biotin supplementation improves hoof strength and decreases the incidence of heel horn junction cracks, sidewail horn cracks and lameness. In one study undertaken by A.Comben 2, 40 horses with a history of chronic foot problems were supplemented with 15mg of biotin per day.
These horses had varying symptoms ranging from brittle 'dished' hooves, to tender crumbling hooves with toe and heel cracks. After five months of treatment, previously sceptical farriers were amazed at the change in the hoof horn of these horses. The feet had developed a better shape and the horn of the hoof walls was thicker and harder. Farriers reported that shoes could be nailed on more satisfactorily and because there was improved hoof growth, there was more opportunity to shape the hooves. These researchers reported that improvements were evident within 3-6 months of biotin supplementation, with continued improvement noted for up to 12 months. However, Comben failed to report that biotin supplementation had little or no effect on some of the horses. It is also reported that biotin supplementation is effective only for horses with hoof defects involving the stratum externurn.3
Professor S. Kempson from the University of Edinburgh undertook a study to investigate hoof horn defects with the use of a scanning electron microscope. The team at Edinburgh found that it was possible to locate the exact origin of the hoof defect in the hoof horn. They discovered that some cracks were located in the stratum externum. Cracks in this layer typically showed a loss of horn material in the outer layer. Other cracks involved the stratum medium and the stratum internum due to a breakdown of structural organisation in these layers.3 On further scrutiny, they realised that horses with defects in the stratum exteruum responded very well to biotin supplementation, whereas cracks that involved the deeper layers benefited most from the addition of protein and calcium to the diet. Most people do not have access to scanning electron microscopy, therefore it is impossible to say where the weakness originates from in the hoof horn. BIOTEX-37 is a complete hoof supplement which will supply all of the nutrients to care for every layer in the hoof horn. BIOTEX-37 is the optimum supplement to use in cases of chronic foot problems.
Calcium
As already mentioned above, Professor Kempson found that horses with defects in the stratum medium and stratum internum responded better to supplementation with a combination of calcium, protein, bioavailable sulphur and biotin, rather than just biotin alone. High levels of protein in the diet favour the uptake of calcium as opposed to phosphorus.
BIOTEX-37 contains calcium and a high level of amino acids, the building blocks of protein. The team at the University of Edinburgh in Scotland concluded that the addition of calcium to the diet of horses, often alleviated hoof problems that did not respond to just biotin alone. "In conclusion, these observations have revealed two types of structural defect in the horn of horses with brittle feet. The second type of defect may fail to correspond to biotin supplementation but will show an improvement with increased calcium and increased protein in the diet"3 Any horse that is fed too much bran or any high phosphorus, low protein diet will inhibit the uptake of this important mineral. Of further consequence, large intakes of bran are associated with an elevated level of phosphorus, which is often a precursor to a sudden outbreak of laminitis.6 The addition of calcium to the diet has both therapeutic and prophylactic properties for laminitis. Therefore BIOTEX-37 is an ideal supplement for horses with chronic foot problems and horses that may be at risk of laminitis, or indeed horses that are recovering from the illness.
BIOTEX-37 contains 7,500mg of calcium carbonate, the level recommended by Professor Kempsom at the University of Edinburgh.
Bioavailable Sulphur: Methionine & MSM
Nutritionists, farriers and veterinarians are now beginning to concur that bio-available sulphur is an extremely important nutrient in the diet of the horse, especially with regard to hair and hoof growth; and integrity of connective tissue. According to the 1989 National Research Council (NRC) publication "Nutrient Requirements of the Horse", the recommended daily allowance (RDA) for sulphur in the horse is 0.15% of the diet dry matter. The horse like other nonruminants must meet its sulphur requirements from bio-available sulphur, therefore both the source and the quality of bio-available sulphur is very important in the equine diet. New sulphur enriched BIOTEX-37 contains two excellent sources of bio-available sulphur; the nutrient Methylsulphonylmethane (MSM) and the essential ammo acid Methionine. Every 20g of BIOTEX-37 contains 2,500mg of MSM and 2,500mg of methionine. Both of these nutrients are easily absorbed sources of dietetic sulphur. The importance of sulphur in relation to improved hoof growth relates to the durability of the keratin sulphate molecule. The keratin sulphate molecule relies on the integrity of it's numerous disulphide bonds made up of the sulphur-bearing amino acids. Methionine is one is these essential amino acids therefore its' inclusion in BIOTEX-37 is fundamental for improved integrity of keratin sulphate. MSM is a nutrient rich in organic sulphur.
MSM provides 34% bioavailable-available sulphur, ensuring that there is adequate sulphur being provided in the diet for optimum hoof growth.
Zinc
The major function of Zinc in the body is to act as a component of many metalloenzymes involved in protein and carbohydrate metabolism. Zinc compounds are the oldest form of therapy for connective tissue disorders. Zinc is essential for DNA and protein synthesis, and is paramount for the healing of damaged epithelial cells. As keratin is largely a proteinaceous material, zinc is an essential ingredient in BIOTEX-37 for the proper healing of hoof horn tissue. In healthy animals, zinc deficiency is related to depressed utilisation of amino acids and sulphur. Therefore, BIOTEX-37 contains a concentrated source (600mg of elemental zinc per 20g) of this essential trace element to promote optimal growth and healing in the hoof horn.
Amino Acids
BIOTEX-37 contains a full spectrum of dispensable and nondispensable amino acids. These nutrients are the building blocks of protein. Their inclusion in BIOTEX-37 is very important for complete healing and improved integrity of keratin sulphate - the main protein in the hoof horn.

Keratinsulfat stellt die strukturelle Komponente in dieser Matrix dar. Keratinsulfat ist ein Mukopolysaccharid und besitzt als solches eine große Affinität für Wassermoleküle. Die Integrität des Keratinsulfats ist fundamental, weil die Wasseraffinität des Keratins den hohen Feuchtigkeitsgehalt im Hufhorn ermöglicht. Die Aufrechterhaltung des Feuchtigkeitsgehalts im Hufhorn ist extrem wichtig. Elastizität und die Fähigkeit, Druck zu absorbieren, sind essenzielle Faktoren in der Verteilung der durch die Fortbewegung auftretenden Erschütterungskräfte. Wenn die Feuchtigkeit im Huf unzureichend ist, wird das Hufhorn zerbrechen. Wie bereits oben erwähnt, besteht das Hufhorn überwiegend aus Hornzellen und Keratinsulfat (ein proteinartiges Material mit einem hohen Schwefelgehalt). Wenn das Hufhorn durch irgendwelche Umstände nicht ausreichend mit hochwertigem Protein, bioverfügbarem Schwefel, Kalzium, Zink oder Biotin versorgt wird, können Unregelmäßigkeiten in der Hufwand auftreten. Dies wird sich in Form von rissigen oder spröden, zerbröckelnden Hufen manifestieren. Das Pferd hat sich als Art gut in der Evolution entwickelt und an Veränderungen angepasst, das heißt natürlich bis zur Einführung des Menschen und des Pferdesports. Seit ihrer Domestikation sind Pferde zur Arbeitsleistung genutzt worden.
Umweltfaktoren, wie z. B. künstliche Arbeits- und Rennbeläge, Straßen, gewisse Einstreu und getreidereiche Rationen, haben alle einen erhöhten Druck auf die Selbsterhaltung der Pferdehufe ausgeübt. In vielen Fällen reicht die Diät des Stallpferdes nicht aus, um den Nährstoffbedarf für die Keratinisierung des überlasteten Hufhorns zu decken. BIOTEX-37 liefert alle wissenschaftlich nachgewiesenen Nährstoffe, die durch das Pferd zur Aufrechterhaltung einer gesunden Hufproduktion benötigt werden.
Topographisch betrachtet ist der Huf in drei Regionen unterteilt: die Zehe, die medialen und lateralen Seitenwände und die Trachten. Die Trachtenwände verlaufen nach innen und bilden die Eckstreben, welche vom Strahl durch die Strahlfurchen getrennt sind. Die Wand ist an der Zehe am dicksten und wird zu den Trachten hin allmählich dünner.
Die Wandstruktur des Hufs
Die Wand ist dazu konzipiert, sowohl den Großteil des Gewichts des Pferdes zu tragen als auch die tieferliegenden Strukturen zu schützen.
Die Eckstreben fungieren als Stütze, um eine Überdehnung und die Kontraktion des Hufs zu regulieren.
Die Sohle bedeckt die weicheren Gewebe und ist etwas konkav, um Halt zu geben und eine Ausdehnung zu ermöglichen.
Der Strahl hilft dabei, Erschütterung zu absorbieren und unterstützt die Zirkulation sowie die Feuchtigkeitsregulation im Huf.
Die Wand ist in drei Schichten unterteilt. Das Stratum externum, das Stratum medium und das Stratum internum. Wenn in der Hufwand ein Riss oder eine Fehlbildung vorliegt, kann die Schwachstelle normalerweise lokalisiert werden, und zwar entweder in der Wandschicht (Stratum externum) oder in den mittleren und inneren Schichten (Stratum medium und Stratum internum). Eine genaue Diagnose kann nur mittels Elektronenmikroskopie und Biopsie erstellt werden. Während es unmöglich ist, mit bloßem menschlichen Auge festzustellen, wovon die Schwachstelle ausgeht, ist die korrekte Diagnose sehr wichtig im Hinblick auf die Ernährungstherapie, die zur Lösung des Problems angewendet werden sollte.
Hufwachstum & Die Jahreszeiten
Die Hufwand an sich ist überwiegend aus keratinisiertem Gewebe zusammengesetzt und stellt so die modifizierte Epidermis der Haut dar. Die innerste, an die Lederhaut angrenzende Hornschicht ist indes eine aus Hornzellen bestehende Wachstumsschicht. Die Zellen aus dieser Schicht teilen sich kontinuierlich, um Hornzellen zu bilden, die der Hornmasse hinzugefügt werden. Das Wachstum erfolgt röhrenförmig von der Oberfläche der aus der Kronlederhaut herausragenden Zotten.
Die Hufwand ist deshalb aufgebaut aus Hornröhrchen, die durch nicht röhrchenförmiges Horngewebe zusammengehalten werden. Die Richtung des Hornwachstums verläuft vom Kronrand abwärts, sodass das Röhrchenhorn nach unten gedrückt wird, um die Wand- und Tragflächen zu bilden: Auf diese Weise befindet sich das jüngste Horn dem Koronarband am nächsten, und das älteste Horn ist auf der Tragfläche der Wand. Diese Methode des Wachstums der Wand erklärt das Vorkommen von Leisten im Horn.
Wenn die Wachstumsrate unregelmäßig ist, ist die Wand von sich abwechselnden Leisten und Vertiefungen umgeben. Der Nährstoffgehalt der Diät, welcher durch die Jahreszeiten beeinflusst ist, ist mitunter ein Einflussfaktor auf die Rate der Hornproduktion.1 Wenn das Pferd im Frühjahr draußen auf der Weide ist, ist das Hufwachstum schneller, üppiger und produziert so die erhabenen „Grasringe“. Während dieser Zeit ist der Nährstoffgehalt der Weide signifikant besser als in den Herbst- und Wintermonaten. Im Winter hingegen ist das Hufwachstum tendenziell vermindert und von geringerem Ausmaß, und folglich können kreisförmige Vertiefungen die Jahreszeit markieren. Die überwiegende Mehrheit der sich im Training befindlichen Pferde hat einen eingeschränkten Zugang zu qualitätvollem, frischem Futter, und deshalb sind Ernährungsmängel nicht unüblich.
Keratinisierung (Hufwachstum)
Eine Fehlernährung wird die physiologischen Prozesse im Körper beeinträchtigen. Diese Ernährungsmängel werden sich auf viele unterschiedliche Weisen manifestieren. Unregelmäßigkeiten in den epidermalen Schichten, wie z. B. der Haare und Hufe, deuten auf Nährstoffdefizite in der Diät des Pferdes hin.
Die Hufwand besteht aus mikroskopischgroßen Röhrchen. Die Räume zwischen diesen Röhrchen sind mit dichtgepackten Hornzellen angefüllt. Auf diese Weise werden die Röhrchen zusammengehalten. Keratin ist das für die Verbindung dieser Zellen und Röhrchen essenzielle Protein.
Diese Struktur ermöglicht dem Horn, sich auszudehnen und Erschütterungen zu absorbieren und reduziert so die Abnutzung der Knochen, Bänder und Sehnen des Vorderfußes. Der Großteil der Hufwand wird aufgrund seines hohen Zellgehaltes durch das Koronarband gebildet. Das Koronarband ist ein Streifen stark vaskularisierten Gewebes, das um den oberen Rand der Wand direkt unterhalb der Haarlinie verläuft und Horn auf ähnliche Weise bildet wie die Haut Haare. Wie Haare besteht Horn hauptsächlich aus Protein. Die Keratinisierung hängt von der normalen Aktivität der Hornzellen ab. Da im Koronorband eine hohe Konzentration der Hornzellen vorliegt, ist der Gesundheitszustand dieses vaskulären Gewebes fundamental. Ernährungsmängel, Traumen, Abrieb und Infektion des Koronarbands sind die wahrscheinlichsten limitierenden Faktoren für die Keratinproduktion im Hufhorn.
Die Keratinisierung ist abhängig von der gesunden Aktivität der Hornzellen, welche zur Bildung dieses „Klebstoffs des Hufhorns” bestimmte Nährstoffe benötigen. BIOTEX-37 wurde speziell formuliert, um einen konzentrierten und ausgewogenen Gehalt dieser Nährstoffe für eine optimale Rate der Keratinisierung bereitzustellen. Dies bedeutet, dass BIOTEX-37 helfen wird, Unregelmäßigkeiten in allen drei Schichten der Hufwand zu kontrollieren und zu korrigieren.

Diese drei Schichten gehen aus dem die Kronlederhaut bedeckenden Epithel hervor. Die äußere Schicht ist eine extrem dünne Schicht aus röhrchenartigem Horn und bedeckt die gesamte Wandoberfläche. Die mittlere Schicht oder Stratum medium besteht aus einer sehr dicken und harten Schicht aus Röhrchenhorn und Zwischenröhrchenhorn. Der proximale Anteil teilt sich unter Bildung des Kronsaums, in welchem die konvexe Kronlederhaut liegt. Er ist pigmentiert, wenn der Huf dunkel ist. Der mittlere Anteil umfasst tatsächlich den Großteil der Hufwand. Die Räume zwischen den Hornröhrchen sind mit Zwischenröhrchenhorn gefüllt. Die innere Schicht oder Stratum internum ist stets nichtpigmentiert und besteht aus vielen keratinisierten primären Epidermislamellen, welche sich vom Kronsaum zur Tragfläche hin ausdehnen.
Die wissenschaftliche Forschung hat festgestellt, dass Hufrisse von einer oder zwei dieser drei Schichten ausgehen. Die Forschung hat zudem gezeigt, dass der Ursprung des Hufdefekts letztendlich die Auswahl der Ernährungstherapie entscheiden sollte. Risse, die vom Stratum externum ausgehen, sprechen sehr gut auf eine Biotinsupplementierung an.2 Risse, die vom Stratum medium und Stratum internum ausgehen, werden besser auf eine Ergänzung der Biotinsupplementierung mit Protein, Kalzium, bioverfügbarem Schwefel und Zink ansprechen.3

Fußpflege
Ein dreifacher Ansatz
1. Ernährung: Alle Pferde, die sich im Training befinden und von denen eine Leistung auf höchstem Niveau erwartet wird, sollten eine ausgewogene Diät erhalten.
Da der Huf und die tieferliegenden Strukturen das Fundament des Pferdes darstellen, sollte ihrer Versorgung eine vorrangige Aufmerksamkeit gewidmet werden. Lahmheit ist die hauptsächliche Ursache einer suboptimalen Leistung des Pferdes. Interessanterweise ist die Fußlahmheit die beim equinen Athleten am häufigsten beobachtete Form der Lahmheit.4 BIOTEX-37 ist das ultimative Ergänzungsfutter für die Hufe, da es essenzielle Nährstoffe zur Versorgung eines jeden Gewebes im Hufhorn liefert.
2. Fußhygiene: Zunächst einmal sollten die Ställe so gestaltet sein, dass sie ein Ablaufen von Urin und Wasser berücksichtigen. Frische Luft sollte in der Stallumgebung eine Priorität sein. Die Wahl der Einstreu ist subjektiv, jedoch ist es wichtig, dass die Einstreu in jedem Fall nicht zu trocken ist, da dies den natürlichen Feuchtigkeitsgehalt des Hufhorns beeinträchtigen wird. Die Einstreu sollte so frisch wie möglich gehalten werden. Dies wird die Anreicherung und Etablierung von endemischen Bakterien und Pilzen in der Stallumgebung reduzieren. Etliche dieser Mikroorganismen haben einen natürlichen Tropismus für das Hufgewebe, z. B. bei Strahlfäule.
3. Hufschmiedearbeit: Ein qualifizierter Hufschmied sollte den Huf stets beschneiden und einen Hufbeschlag durchführen. Der Hufschmied hat einen gewaltigen Einfluss auf die Balance des Fußes. Die Herrichtung des Hufs, die Art des Beschlags und seine Platzierung werden alle die Verlagerung der Kraft auf den Vorderfuß und die angrenzende Gliedmaße beeinflussen. Viele Hufdefekte haben multifaktorielle Ursachen. Ein inkorrekter Beschlag und Vernachlässigung kann einer der prädisponierenden Faktoren für Erkrankungen, eine Störung der Hufhornfunktion sowie rissige und spröde Hufe sein. Eine sachgerechte Hufpflege in Verbindung mit hygienischen Haltungsbedingungen und einer Fütterung von BIOTEX-37 ist der beste Ansatz, um die Hufgesundheit und ein optimales Hufwachstum zu gewährleisten.
Wie wirkt BIOTEX-37?
Biotin
Biotin gehört zur Familie der B-Vitamine, wurde geschichtlich jedoch als Vitamin H bezeichnet. Biotin ist ein schwefelhaltiges Vitamin. Grünfutter ist die beste Quelle für Biotin in der Diät. Pferde werden zudem mit Biotin und anderen B-Vitaminen als Nebenprodukte der bakteriellen Aktivität im Dickdarm versorgt. Die moderne Haltung des Leistungspferdes bedingt jedoch lange Zeitperioden mit wenig oder keinem Zugang zu einer Weide mit guter Qualität.
Pferde, die sich im Training befinden und denen eine getreidereiche und grünfutterarme Diät gefüttert wird, leiden oftmals an einem B-Vitaminmangel. Dieser ist bedingt durch die Tatsache, dass die schnelle Stärkeverdauung den natürlichen pH im Zäkum absenkt, wodurch als Nettoergebnis eine Verminderung der B-vitaminbildenden Bakterien im Dickdarm resultiert. Deshalb sind alle Stallpferde im Training einem potenziellen Risiko eines Biotinmangels ausgesetzt.
Pferde, die der Gefahr eines Biotinmangels ausgesetzt sind:
• Mangel an Biotin in der Diät.
• Verlust der mikrobiellen Aktivität im Darm, durch antibiotische Therapie oder eine veränderte Darmmotilität.
• Trächtige oder laktierende Stuten oder betagte, geriatrische Pferde.
• Angeborene Erkrankungen des Biotinmetabolismus.
Biotinsupplementierung
Die empfohlene Tagesdosis (Recommended Daily Allowance, RDA) für Biotin für ein 500 kg-Pferd beträgt 2 mg pro Tag.5 Die Forschung hat jedoch gezeigt, dass eine erhöhte Biotinsupplementierung der Diät von Pferden mit Hufproblemen zu einem verbesserten Hufwachstum und insgesamt zu einer verbesserten Integrität des Hufs führt. Jede 20 g BIOTEX-37 enthalten 37.5 mg Biotin. Dies ist ein konzentriertes Niveau, das die Biotinabsorption im Dünndarm maximiert. Die Biotinsupplementierung wird die Reparatur von Hufdefekten erhöhen und helfen, ihr Wiederauftreten zu verhindern. Es gibt stichhaltige Hinweise aus der Wissenschaft, dass eine Biotinsupplementierung die Widerstandskraft des Hufs verbessert und die Inzidenz von Rissen am Übergang zum Ballenhorn, des Seitenwandhorns sowie von Lahmheiten reduziert. In einer durch A. Comben2 durchgeführten Studie wurde 40 Pferden mit einer Vorgeschichte chronischer Fußprobleme eine Supplementierung von 15 mg Biotin pro Tag verabreicht.
Diese Pferde wiesen unterschiedliche Symptome, von spröden, konkaven Hufen bis weichen, zerbröckelnden Hufen mit Zehen- und Ballenrissen, auf. Nach einer fünfmonatigen Behandlung waren die vorher skeptischen Hufschmiede erstaunt über die Veränderung des Hufhorns dieser Pferde. Der Fuß hatte eine bessere Form entwickelt, und das Horn der Hufwände war dicker und härter. Die Hufschmiede berichteten, dass die Eisen zufriedenstellender angenagelt werden konnten, und da das Hufwachstum verbessert war, gab es mehr Möglichkeiten, die Hufe zu formen. Diese Forscher berichteten, dass die Verbesserungen innerhalb von 3-6 Monaten der Biotinsupplementierung offensichtlich waren und bis zu 12 Monate lang eine kontinuierliche Verbesserung beobachtet wurde. Comben hat indes nicht berichtet, dass eine Biotinsupplementierung bei einigen der Pferde einen geringen oder keinen Effekt hatte. Es wurde zudem beschrieben, dass eine Biotinsupplementierung nur wirksam bei Pferden mit Hufdefekten des Stratum externum ist.3
Professor S. Kempson von der Universität Edinburgh unternahm eine Studie zur Untersuchung von Hufhorndefekten unter Verwendung eines Rasterelektronenmikroskops. Das Team in Edinburgh stellte fest, dass es möglich war, den exakten Ursprung des Hufdefekts im Hufhorn zu lokalisieren. Sie entdeckten, dass einige Risse im Stratum externum lokalisiert waren. Die Risse in dieser Schicht wiesen typischerweise einen Verlust an Hornmaterial in der äußeren Schicht auf. Andere Risse betrafen das Stratum medium und das Stratum internum aufgrund eines Zerfalls der strukturellen Organisation in diesen Schichten.3 Nach weiteren Untersuchungen erkannten sie, dass Pferde mit Defekten im Stratum externum sehr gut auf eine Biotinsupplementierung ansprachen, wohingegen Risse, die die tieferen Schichten betrafen, am meisten von einer Ergänzung der Diät mit Protein und Kalzium profitierten. Die meisten Leute haben keinen Zugang zur Rasterelektronenmikroskopie, und folglich ist es nicht möglich, zu sagen, wo im Hufhorn die Schwachstelle ihren Ursprung hat. BIOTEX-37 ist ein komplettes Hufergänzungsmittel, das alle Nährstoffe zur Versorgung jeder Schicht im Hufhorn liefern wird. BIOTEX-37 ist das optimale Ergänzungsmittel zur Anwendung bei chronischen Fällen von Fußproblemen.
Kalzium
Wie bereits oben erwähnt, fand Professor Kempson heraus, dass Pferde mit Defekten im Stratum medium und Stratum internum besser auf eine Supplementierung mit einer Kombination aus Kalzium, Protein, bioverfügbarem Schwefel und Biotin ansprachen als mit Biotin alleine. Hohe Proteingehalte in der Diät begünstigen die Aufnahme von Kalzium im Gegensatz zu Phosphor.
BIOTEX-37 enthält Kalzium und einen hohen Gehalt an Aminosäuren, die Bausteine der Proteine. Das Team an der Universität Edinburgh in Schottland kam zu dem Schluss, dass eine Ergänzung der Diät der Pferde mit Kalzium oftmals Hufprobleme linderte, die alleine auf Biotin nicht ansprachen. „Schlussendlich haben diese Beobachtungen zwei Typen eines strukturellen Defekts im Horn von Pferden mit spröden Füßen aufgezeigt. Der zweite Defekttyp spricht mitunter nicht auf eine Biotinsupplementierung an, aber er wird eine Verbesserung infolge einer Erhöhung des Kalzium- und Proteingehaltes in der Diät zeigen."3 Jedes Pferd, dem zu viel Kleie oder eine phosphorreiche und proteinarme Diät gefüttert wird, hemmt die Aufnahme dieses wichtigen Mineralstoffs. Große Aufnahmemengen an Kleie sind folglich weiterhin assoziiert mit einem erhöhten Phosphorspiegel, was oftmals ein Wegbereiter eines plötzlichen Hufreheausbrauchs ist.6 Eine Ergänzung der Diät mit Kalzium hat für die Hufrehe sowohl therapeutische als auch prophylaktische Eigenschaften. Deshalb ist BIOTEX-37 ein ideales Ergänzungsmittel für Pferde mit chronischen Fußproblemen und Pferde, die rehegefährdet sein könnten oder sich sogar von dieser Erkrankung erholen.
BIOTEX-37 enthält 7,500 mg Kalziumkarbonat, den durch Professor Kempson an der Universität Edinburgh empfohlenen Gehalt.
Bioverfügbarer Schwefel: Methionin & MSM
Ernährungswissenschaftler, Hufschmiede und Tierärzte beginnen nunmehr übereinzustimmen, dass bioverfügbarer Schwefel ein extrem wichtiger Nährstoff in der Diät des Pferdes ist, besonders im Hinblick auf das Haar- und Hufwachstum und die Integrität des Bindegewebes. Gemäß der 1989 erschienen Publikation „Nährstoffbedarf des Pferdes" des Nationalen Forschungsrates (National Research Council, NRC) beträgt die empfohlene Tagesdosis (RDA) für Schwefel 0.15 % der Trockensubstanz in der Diät. Wie andere Nichtwiederkäuer muss das Pferd seinen Schwefelbedarf durch bioverfügbaren Schwefel decken, und demzufolge sind sowohl die Quelle als auch die Qualität des bioverfügbaren Schwefels in der Diät des Pferdes sehr wichtig. Das neue, mit Schwefel angereicherte BIOTEX-37 enthält zwei exzellente Quellen bioverfügbaren Schwefels: den Nährstoff Methylsulfonylmethan (MSM) und die essenzielle Aminosäure Methionin. Jede 20 g BIOTEX-37 enthalten 2,500 mg MSM und 2,500 mg Methionin. Diese beiden Nährstoffe sind einfach absorbierbare Quellen diätetischen Schwefels. Die Bedeutung des Schwefels für das verbesserte Hufwachstum steht im Zusammenhang mit der Strapazierfähigkeit des Keratinsulfatmoleküls. MSM liefert zu 34 % bioverfügbaren Schwefel und gewährleistet so, dass die Diät ausreichend Schwefel für ein optimales Hufwachstum bereitstellt.
Zink
Die hauptsächliche Aufgabe von Zink im Körper ist seine Funktion als Bestandteil vieler am Protein- und Kohlenhydratmetabolismus beteiligter Metalloenzyme. Zinkverbindungen sind die älteste Therapieform für Bindegewebserkrankungen. Zink ist essenziell für die DNA- und Proteinsynthese und es hat Priorität für die Heilung geschädigter Epithelzellen. Da Keratin größtenteils ein proteinartiges Material ist, ist Zink in BIOTEX-37 ein essenzieller Bestandteil für die einwandfreie Heilung des Hufhorngewebes. In gesunden Tieren steht ein Zinkmangel mit einer herabgesetzten Nutzung der Aminosäuren und des Schwefels in Zusammenhang. BIOTEX-37 enthält deshalb eine konzentrierte Quelle (600 mg elementaren Zinks pro 20 g) dieses essenziellen Spurenelements, um ein optimales Wachstum und eine optimale Heilung des Hufhorns zu unterstützen.
Aminosäuren
BIOTEX-37 enthält ein komplettes Spektrum essenzieller und nichtessenzieller Aminosäuren. Diese Nährstoffe sind die Bausteine der Proteine. f) Feuchtigkeitsgehalt – Keratinsulfat besitzt eine große Affinität zu Wasser. Das Vorhandensein von Wasser ist sehr wichtig für die Niederlegung und die natürliche Elastizität des Hufhorns.

Clinical testing results confirmed studies showing that icariin is a PDE5 inhibitor treat erectile dysfunction, boosting libido and increase desire.
Prof. Dell'Agli and his colleagues tested the four plants in vitro to see how efficient they were at inhibiting PDE5. Just one - Epimedium brevicornum, also known as horny goat weed - had an effect. This confirmed previous studies showing that icariin, a compound found inside the horny goat weed, is a PDE5 inhibitor boosting libido, increase sexual desire, and treat erectile dysfunction.
The activity of plant extracts and individual compounds against human recombinant PDE5A1 is shown in Figure S1 (Supporting Information). Cinnamaldehyde, icariin (1), and ferutinin were considered as the putative active principles of C. cassia, E. breVicornum, and F. hermonis, respectively, since the compounds represent the most abundant secondary metabolites of those species. Only E. breVicornum and icariin (1) strongly inhibited PDE5A1 (-80% and -72%, respectively), whereas the other test materials were much less active (-15 to -23%). Inhibition by cinnamalde-hyde (-16%) and ferutinin (-7%) was not significant.
The medicinal plants tested in the present study had a reputation for aphrodisiac effects and therefore represented the start of a screening program to search for compounds to be developed as a new natural drug alternative to sildenafil. The observation that only E. brevi-cornum and its active principle 1 inhibited PDE5 in a significant manner, in agreement with previous results,17-20 suggests that the other plant extracts may interfere with erectile function through mechanisms other than PDE5 inhibition.
Compound 1 was a good PDE5 inhibitor (IC50 of 5.9 μM), but required improvement in order to have equivalent potency to sildenafil, which gave an IC50 of 75 nM. The inhibitory effects of icariin derivatives 2-6 on PDE5A1 is shown. Since aglycons might be expected to possess higher activity than the corresponding glycosides, the first general structural modification to 1 was the removal of one or both of the sugar moieties at the hydroxyl groups at positions C-3 and C-7 of the flavone scaffold.
Enzymatic hydrolysis of 1 with cellulase and naringinase allowed the partial or total removal of the sugar moieties, respectively.
The last modification to 1 was the replacement of one or both sugar moieties with the hydroxyethyl side chain, representing a simplification of the sugar residue. The substitution for Glc by a hydroxyethyl group at C-7 gave 7-(2-hydroxyethyl)-3-O-rhamno-sylicariin (4), which was less potent than 2 (IC50 363 vs 156 nM, respectively). When both hydroxyls at C-3 and C-7 were derivatized with hydroxyethyl moieties, as in 3,7-bis(2-hydroxyethyl)icaritin (5), PDE5A1 was potently inhibited, with an IC50 of 74 nM, almost identical to that of sildenafil (75 nM).
The selectivity against human PDE5A1 was investigated by testing all compounds against human PDE6C, at concentrations 10-fold higher than their PDE5A1 IC50 values. Compounds 1 and 4 inhibited PDE6 activity (45% and 86%, respectively), while 2, 3, 5, and 6 were inactive.
For 5, the best PDE5 inhibitor among the icariin derivatives tested, concentration inhibition curves for PDE6 and cAMP-PDE were performed. The PDE6C/PDE5 IC50 ratio was 418 for 5 and 2.2 for sildenafil, while the cAMP-PDE/PDE5 IC50 ratio was 1300 for 5 and 367 for sildenafil. These results indicate that the selectivity of 5 for PDE5 was improved with respect to that of sildenafil. Compound 5 was not cytotoxic for human fibroblasts even at the highest concentration tested (100 μM). Thus, the inhibitory potency of 5 was 80-fold higher that of the parent compound icariin.
Preparation of Icariin
A solution of icariin (1) (526 mg) in DMSO (1 mL) was added dropwise for 72 h to a Na acetate-buffered hydroalcoholic solution at 37 °C (0.25 M, pH 5.0, in EtOH/H2O, 30: 70) (50 mL) containing naringinase (207 mg). The obtained suspension was allowed to stir at 37 °C for 7 days.
Then, a further amount of naringinase (97 mg) was added and the mixture was stirred under the same conditions for a further day. EtOH was removed by evaporation and the aqueous suspension was filtered under vacuum and dried.
The residue obtained was washed with H2O and dried to give icaritin (3, 290 mg; purity 95%) as a yellow powder. The mother liquors were diluted with H2O and extracted with EtOAc (2 × 200 mL). The organic phase was dried over anhydrous Na2SO4 and evaporated under reduced pressure to afford an additional amount of 3 (20 mg); quantitative yield.

Die Beobachtung, dass nur E. brevicornum und sein aktiver Wirkstoff 1 PDE5 in signifikanter Weise hemmten, stimmt mit früheren Ergebnissen (17-20) überein und legt nahe, dass die anderen Pflanzenextrakte die Erektionsfunktion nicht durch PDE5-Hemmung, sondern durch andere Mechanismen beeinträchtigen.
Verbindung 1 war ein guter PDE5-Hemmer (IC50 von 5,9 µm), erforderte jedoch Verbesserungen, um genauso wirksam wie Sildenafil (C50 von 75 nm) zu sein. Die letzte Änderung von 1 war der Austausch einer oder beider funktioneller Zuckergruppen mit einer Hydroxyethyl-Seitenkette, was einer Vereinfachung der Zuckerreste entsprach. Der Austausch von Glc durch eine Hydroxyethyl-Gruppe an C-7 ergab 7-(2-Hydroxyethyl)-3-O-Rhamno-Sylicariin (4), das weniger stark wirksam war als 2 (IC50 363 vs. 156 nM). Wenn beide Hydroxyl-Gruppen an C-3 und C-7 mit Hydroxyethyl-Gruppen derivatisiert wurden, wie in 3,7-Bis(2-Hydroxyethyl)Icaritin (5), wurde PDE5A1 auf wirksame Weise gehemmt; der resultierende IC50-Wert war mit 74 nM fast identisch mit dem von Sildenafil (75 nM).

Für 5, den besten PDE5-Inhibitor unter den getesteten Icariin-Derivaten, wurden Konzentrations-Hemmkurven für PDE6 und cAMP-PDE durchgeführt. Das PDE6C/PDE5IC50-Verhältnis betrug für 5 418 und für Sildenafil 2,2, während das cAMP-PDE/PDE5-IC50-Verhältnis für 5 bei 1300 und für Sildenafil bei 367 lag. Diese Ergebnisse weisen darauf hin, dass die Empfindlichkeit von 5 für PDE5 in Bezug auf Sildenafil verbessert wurde. Aufbereitung des Icariins
Eine Lösung von Icariin (1) (526 mg) in DMSO (1 ml) wurde tropfenweise für 72 h zu einer 37 °C warmen Natriumacetat-gepufferten, wässrig-alkoholischen Lösung (0,25 M, pH-Wert 5,0, in EtOH/H2O, 30: 70) (50 ml) mit Naringinase (207 mg) gegeben. Die erhaltene Suspension wurde 7 Tage lang bei 37 °C gerührt.
Dann wurde eine weitere Menge Naringinase (97 mg) hinzugefügt, und das Gemisch wurde unter den gleichen Bedingungen einen weiteren Tag lang gerührt. Das EtOH wurde durch Eindampfen entfernt, und die wässrige Suspension wurde unter Vakuum filtriert und getrocknet.
Der gewonnene Rückstand wurde mit H2O gewaschen, getrocknet und ergab Icaritin (3290 mg; Reinheit 95 %) als ein gelbes Pulver. Die Mutterlaugen wurden mit H2O verdünnt und mit EtOAc extrahiert (2 x 200 ml). Die organische Phase wurde über wasserfreiem Na2SO4 getrocknet und unter vermindertem Druck verdampft und ergab so eine zusätzliche Menge von 3 (20 mg); quantitative Ausbeute.

Charlotte
Charlotte lives in France together with her parents and her older sister. She was born with a genetic defect and had severe respiratory problems after birth. At the age of four months she lost weight, so she got a nasogastric tube, followed by the insertion of a PEG button. After being in touch with Valérie of the NoTube team, the family decided to participate in the tube weaning course in Mayenne in 2013. The 3.5 year old girl made great progress regarding her oral intake and after only a few days in the program, she was able to sustain herself entirely by oral means. Even her PEG button could be removed in Mayenne, a fact that makes little Charlotte proud to this day. Since that time, Charlotte eats completely orally and the PEG button stayed in Mayenne altogether.

Charlotte
